Sofia er en New York parterapeut, som selv i årevis har simuleret orgasmer overfor sin mand Rob. En dag får hun besøg af bøsseparret James og Jamie, som overvejer at åbne deres forhold for flere seksuelle partnere. Sofia mister selvkontrollen midt i terapien og fortæller om sine egne problemer. De opfordrer Sofia til at besøge Shortbus - en lukket klub i Brooklyn hvor søgende sjæle finder et kunstnerisk, musisk, politisk og seksuelt frirum.
