Jake er død, men Elwood Blues lever i bedste velgående, og efter 18 år i fængsel har han kun en ting i hovedet: at genskabe sit band "The Blues Brothers". Det lykkes Elwood at samle bandet, og han får en 10-årig og en bartender som nye kolleger og forsangere. Men det lykkes ham også at få hele Illinois' politistyrke, FBI, den russiske mafia og en fascistisk militsgruppe i hælene, mens bandet bevæger sig mod den afgørende musikkonkurrence.
